What are the foundational concepts of molecular biology techniques for beginners?

As a new graduate student, I find I can perform techniques like PCR or cloning by following steps, but I lack the deep understanding to troubleshoot or design experiments independently. I want to move beyond being a technician and become a scientist who comprehends the 'why' behind each step in these fundamental tools.

From my experience mentoring students, the shift from following steps to understanding comes from grasping a few key principles. First, deeply internalize the Central Dogma the precise flow of information from DNA to RNA to protein as every major technique manipulates this pathway. Second, understand the specific base-pairing rules of nucleic acid hybridization, which is the foundation for techniques from PCR to sequencing. Finally, learn the basic kinetics and fidelity of the enzymes you use daily, like polymerases and restriction enzymes. When you see a protocol, you should be able to map each step back to controlling one of these biochemical principles.
