How can RNA expression be controlled in experimental biology?

In my PhD work on gene function, I need precise temporal and quantitative control over specific transcripts. I'm familiar with some tools like RNAi but am looking for a consolidated, experienced overview of the current reliable toolkit, including their best-use cases and common pitfalls to avoid.

Based on my experience, control depends on your goal. For robust knockdown, I've found synthetic siRNA or shRNA vectors are workhorses, though watch for off-target effects. For complete transcriptional repression, CRISPRi is now my go-to for its specificity. Conversely, to induce expression, I recommend tetracycline- or doxycycline-inducible systems; they offer clean temporal control. The key is pairing the method with your experimental timeline and required specificity, always including multiple controls to confirm the observed effect is genuine.
