Advantages of thin-paper chromatography over paper chromatography


I want to separate a set of mixtures. I wanted to use thin-paper chromatography for this purpose. But my friend asked me to use paper chromatography. Why use thin-paper chromatography over paper chromatography?

Yes your friend is right. Use thin-layer chromatography. This is because 1. Development chamber design In thin-paper chromatography you need not suspend the plate from a rod on top of the development chamber as in paper chromatography. Instead it can be easily placed in a slanting position with the bottom edge placed on the chamber base. 2. Sample volume In thin-paper chromatography the quantity of sample required and applied is very small and can be applied using automated sample dispensers. 3.Choice of reagents Coated plates which are in thin layer chromatography has the potential to withstand corrosive spray reagents which is not possible in paper chromatography. Corrosive spray reagents deteriorate the sample spots.

Your friend is right Yashti. It is advisable to use thin-paper chromatography instead of paper chromatography. This is because paper chromatography instruments do not have much automation when compared to that of thin layer chromatography tools. Thin-paper chromatography tools have constant volume sample dispenser, autosampler, documentation and camera for record of separation of mixture.

Both Yashti and Vishwas are right. But the main advantage of thin-paper chromatography is its rigid support. The cellulose paper support in paper chromatography is flexible which is not the case in thin-paper chromatography. Due to support rigidity in thin-layer chromatography, there is less diffusion.
