I would suggest you to go with insertion of foreign DNA fragments technique.     In this method the cDNA is isolated above or obtained from gene bank will be fragmented     utilising the specific restriction enzyme to produce specific cohesive ends.     The cloning vector will be treated with restriction enzyme to generate cohesive ends.

Go with insertion technique. But remember, you must add termini single stranded DNA sequence complementary to a tract of DNA.

I had used isolation DNA method. I would suggest you to use the same. The DNA of interest,     must be genomic DNA or synthetic DNA. The synthetic DNA of interest if included in a particular restriction fragment,     you can isolate it from gel after electrophoresis. If not complementary DNA fragment will be prepared directly by using mRNA.

Choose isolation DNA approach. For details about the steps refer http://www.sfu.ca/bisc/bisc-429/DNA1.html.
