Yes you can definitely use STR technique to examine the individual areas in DNA. In fact it is the best technique.

STR is the only technique that can be used to examine the individuals regions of DNA. it enables you to determine the difference for specific DNA regions in one person vs another, thereby allowing you to differentiate between two individuals.

Well, STR process includes extraction of nuclear DNA from cells. Particular regions of the extracted DNA is then amplified using PCR technique. 
After amplification process, use gel electrophoresis and figure out how many repeats of the STR sequence exist. 
Use fluorescent dyes or silver staining and obtain the required visual assessment of the DNA.

Mukesh, as far as I know there is no alternatives to STR analysis. This process involves 5 steps.
1. DNA isolation
2. PCR amplification
3. Gel electrophoresis
4. STR profile generation
5. Cell line authentication
