Embryo culture involves isolation of an embryo from a given sample for in vitro growth.  Embryo culture involves mature or immature embryo. Mature embryos used for culture are obtained from ripe seeds.  Immature embryo involves immature embryos from unripe or hybrid seeds.

For embryo culture, the ovule, fruit or seed from which the embryo is to be obtained will be sterilised,  and hence you need not sterilise the embryo once again. Salt sucrose is used to offer the embryo with sufficient nutrients.  The culture is enriched with inorganic, organic compounds, inorganic salts and growth regulators.

Organ culture involves isolation of an organ for in-vitro growth. This type of culture involves various methods such as 1.Plasma clot method – This method involves use of a clot that is composed of chick embryo extract in a watch glass.  This method is used for studying morphogenesis in embryonic organ rudiments and actions of various hormones, vitamins and tissues. 2.Raftmethod – The explant will be placed on a raft of lens paper and floated on a serum in a watch glass. 3.Grid method – This method involves perforated stainless steel sheet on which targeted tissue is placed before placing it  in a culture chamber consisting of fluid medium.
