Cloning method over PCR for producing copies of DNA


I wanted to use PCR for my study and produce copies of DNA. But my friend suggested me to use cloning method rather than going for PCR method. Is there any difference between the two methods? Also what are the advantages of cloning method over PCR?

I agree with your friend. It is wise to use gene cloning method. This is because all the polymerases will include error rates for including wrong bases. These errors occur near the primers. But this is not in the case of cloning method.

Yes, of course there are differences between gene cloning and PCR methods. The PCR process breaks down DNA by heating, unwinds DNA double helix into single strands. Once the strands are separated, DNA polymerase reads the nucleic acid sequence and generates a duplicate strand of DNA. Whereas DNA cloning isolates the source & vector DNA and then cuts the two DNA.

Sumitra, if you want to generate multiple copies, it is better to go with PCR process itself. You can easily create duplicate copies within few seconds.

Thank you all for the suggestions. Will go with gene cloning method as I cannot deal with error rates.
