Aseptic plant preparing method vs sterilization procedure


Which is the best method for plant tissue culture? Am confused among technique of preparing aseptic plants and sterilization procedure.

I would suggest you to go with aseptic plant method. The surface sterilization of plant tissue causes deleterious effect as the sterilants are toxic chemicals. Seeds are cannot resist such deleterious effect due to the presence of seed coat. Inorder to avoid sur­face sterilisation of plant tissue and seeds are sur­face sterilised and then seeds in culture give rise to an aseptic seedlings.

Go with sterilization method. This method not just involves sterilization of plant materials but also sterilization of non-living materials like glass tube, nutrient medium etc used during the experiment.

Go with sterilization procedure. The culture medium consisting of sugar support the growth of microorganisms. Hence if they come in contact with cellular form or in spore form, the micro-organisms grow faster than the higher plant tissue and kills the tissue. Hence, the surface of plant tissue including nutrient medium should be sterilized.
