Haematoxylin-Safranin Method vs Safranin-Fast Green Method


Which is the best approach that can be used for the purpose of double staining in botany? Haematoxylin-Safranin Method or Safranin-Fast Green Method?

You can use Haematoxylin-Safranin method. In this method keep the sections in Delafield haematoxylin for around five minutes and eliminate the excess of stain using water. Next wash the section with ammonia till strain turns blue. Now stain the section with safranin and wash them with glycerine for removing excess of stain and then mount in glycerine.

According to me Safranin-Fast Green method is the best method. Here keep the section to be stained in safranin for five min­utes and wash it with water. Check only if the thick-walls are strained. Excess of stain is removed by acid alco­hol and wash the section with water to remove traces of acid. Next stain the section with fast green for few seconds. Wash the section with glycerine and then mount in a drop of glycer­ine

You can use Safranin-Fast Green method or Safranin-Aniline Blue method. In Safranin-Aniline Blue method just replace fast green with aniline blue. Remaining steps are the same as Safranin-Fast Green method.
