Negative vs simple staining technique in Botany


Hello all. I’m conducting my research in Botany. I want to colour the bacteria. Which is the best method for the same among negative staining and simple staining method? Any suggestions? Also, explain the procedure briefly.

You can go with negative staining if you want to stain bacteria or yeast. This method is the best for quicker observation of the morphology of yeast or bacteria this tech­nique is applied. All you have to do is, place a drop of bacterial suspension on a clean slide. Next, about 1 ml of suspen­sion must be slide spread (by a needle) and dried in air. This is followed by adding a few drops of nigrosin dye solution on the dry smear and is spread on the slide. Once the smear with dye is dried you can examine the sample.

According to me it is better to go with simple staining method rather than negative staining. This method is specifically meant for bacteria.

Yes, I agree with Zeba. Simple staining approach is better than negative staining. Here, transfer a drop of a bacterial suspension to the centre of the clean slide and spread uniformly. Next allow the smear to dry and fix it by passing over a flame. Place the slide on the end of a petri dish and cover the smear using carbol fuchsin stain. Allow the stain to react and then wash the slide with water. Once it’s dried you can observe the sample.
