For determination of size homogeneity you can use gel filtration. Gel filtration let’s you detect small changes in the 
size of detergent-protein complexes especially when different detergent-proteins are compared. 
Gel filtration is used to give an indication of the suitability of various detergents for a particular protein. 
Therefore separation of gel filtration is an important tool for qualifying the membrane protein preparation.

 if you have 100% amino acid sequence identity then the proteins can be said to be homogenous

 Two proteins can be identical only when the structure, function and their sequences are similar.
